Methylene Blue #

• Aromatase inhibitor.
• 5-HT1A receptor agonist?
• A reversible MAO-AI in miligram dosages and a potent anti-inflammatory. Neuroprotective and energy-boosting. Doesn’t increase serotonin though.
  • It’s used for TBI for cerebral Vasoconstriction (probably due to serotonin from bein MAO-AI?)
• This guy claimed it made him fat RPF
• Can help with SIBO? RPF
• Functions as an electron donor by receiving electrons from Complex I and bringing them to Cytochrome C.
  • Used to treat methemoglobinemia (caused by iron in Hemoglobin being converted from Fe2+ to Fe3+. MB donates, converting it back to Fe2+.)
• NOS inhibitor. via several mechanisms.
  • As such, it’s a Guanylate Cyclase inhibitor.
  • As such it raises blood pressure quite reliably; it is a vasopressor in medical settings.
  • These tend to be antidepressant.
• Lowers Cortisol, Prolactin, Estrogen
• Antimicrobial; kills Candida
• Hard on the renal system?
• Reverses fatty Liver
  • SIRT1 activation by methylene blue, a repurposed drug, leads to AMPK-mediated inhibition of steatosis and steatohepatitis
• Like caffeine, MB absorbs ~20% the first hour, then the rest gradually over 24-48 hours.
• Might produce Free radicals; Peat is now against it
• AChEi? High IC50
  • The antagonism of acetyl choline by methylene blue (1926) paralyzes the Vagus Nerve
• Methylene blue inhibits the function of α7-nicotinic acetylcholine receptors noncompetitive.
• Toxicology and carcinogenesis studies of methylene blue trihydrate (Cas No. 7220-79-3) in F344/N rats and B6C3F1 mice (gavage studies) dose is high as fuck. Gave them cancer etc.
• Ehrlich treated Malaria with it.
• [Protective Role of Methylene Blue in Alzheimer’s Disease via Mitochondria and Cytochrome c Oxidase]
  • The key cytopathologies of AD include decline in cytochrome c oxidase (complex IV), mitochondrial dysfunction, energy hypometabolism, abnormal iron homeostasis, oxidative stress, dimerization of AβPP, and synaptic dysfunction.
• [Methylene blue activates the PMCA activity and cross-interacts with amyloid β-peptide, blocking Aβ-mediated PMCA inhibition]
  • PMCA = plasma membrane Calcium ATPase is inhibited by Amyloid β, and MB is able to block/rescue this by binding to PMCA on the C terminus. Aβ al.so somehow increases MB’s affinity for it, too.
