Niacinamide #
Another no-flush form of supplemented niacin is Inositol hexanicotinate. It increases cell membrane permeability.

Readily converts into the bioactive forms of niacin: NAD+, NADH, NADP, and NADPH.

Lowers temperature and stress hormones.

Can lower free fatty acids. Inhibits Lipolysis/Fatty Acid Oxidation at least in large doses.

- Upregulates liver X receptors.
- NAD+ serves as a substrate for the synthesis of cADPR, used in calcium signalling, important for Neuroplasticity, especially in the hippocampus. However, a study showed that decreased niacin intake and cADPR levels enhanced spatial learning in adult male rats, and when nicotinamide (4g/kg (epic 45g HEQ)) was supplemented, it decreased. R

Lowers Blood Glucose, probably via lowering stress hormones and gluconeogenesis. Debatable if it’s due to any burning of glycogen.

Excess nicotinamide increases plasma Serotonin and Histamine levels #
- Depletes the body’s Methyl Group pool.
- 100mg oral.
- Urinary N-methyl-2-pyridone-5-carboxamide and N-methylnicotinamide:

Supplementation #
- Can go anywhere from 25-1000mg. The 1g+ dosages have benzodiazepine-like anxiolytic effects. Much higher, and it converts into NAD too quickly, and can have liver-damaging effects. It depletes Methyl Groups to some degree. Pairing it with betaine 1:1 can help with all that.
- High doses (1-1.5g) help with diabetes. Should be taken early, while niacin should be taken at dinner.
