WNT (“Wingless-type”) signaling pathway #
- Canonical pathway:
- Also see
- Also see
- Wnt/Ca2+ pathway: Gq?->Phospholipase C -> etc. -> NFAT, CREB
- JNK pathway: DV1 -> RhoA->ROCK and Rac1->JNK->TGF-β
- Since ROCK is a negative regulator of PTEN (and IRS-1) this is anti-mtor
- WNT1->WISP1->PI3K->AKT->…
In humans, there exists WNT1-16, with some subtypes all the while.
-
WNT10A/B are implicated in oncogenesis. B is implicated in adipogenesis, coronary vessel formation, and attenuating pathological fibrosis.
-
WNT4 implicated in very high Intelligence.
- Growing evidence suggests that downregulation of Wnt signaling could be involved in the cognitive decline associated with aging and also with the physiopathology of Alzheimer’s disease (AD). R

Reduces Fibroblast proliferation, probably through antagonizing TGF
-
- By inducibly deleting β-Catenin globally in skin epithelia, only in hair follicle stem cells, or only in interfollicular epidermis, and comparing the phenotypes with those caused by ectopic expression of the Wnt/β-catenin inhibitor Dkk1, we show that this pathway is necessary for hair follicle stem cell proliferation
- β-Catenin is not required within hair follicle stem cells for their maintenance, and follicles resume proliferating after removal of ectopic Dkk1, indicating persistence of functional progenitors.
