NSI-189 (4-benzylpiperazin-1-yl) #

A novel drug, with an epic history. Neuralstem received virtually unlimited funding from DARPA, and it was selected out of 10,000 other compounds for optimal increase in hippocampal volume. It’s unknown what receptors it binds to but as one would assume it raises Neurotrophic Factors. Not a good generic antidepressant, apparently, but works amazingly for some people.

Mechanism #

Pretty much unknown but it’s speculated to be adrenergic somehow.

  • DeltruS: Stacks extremely well with meditation type stuff such as qigong, wim hof, vipassana. Side effects: Nerve twitches, Sensations like pressure and tingling in specific areas, manic days (they seem to have stopped recently?), light inflammation, insomnia if taken in the morning but not at night, usually from thinking too much, typing very long things without paying attention to time or surroundings, sometimes obsessing with trains of thoughts (not in a bad way like depression, more of like poor task switching). Super clean like nothing in the mind is hidden from you. Doesn’t help at all with motivation or memory. - 25 mg every other day. I’ve had more ideas on NSI-189 than I’ve had in my entire life.
  • MisterYouAreSoDumb: Very hyperemotional. Feeling sad for no reason. Mitigate with tianeptine.
  • Very sedating for the first week, which caffeine mostly mitigated - then it became stimulating. *In my previous cycles (thiry-day) I was lethargic, deeply emotional, yet detatched and dreamy. It initially reminded me of Memantine. This cycle, however, has been completely different. I have been rather assertive, quiet, energized, deeply introspective, not very emotional at all, stoic, and curt. I feel like a Spartan. For what it is worth, my previous cycles were phosphate and this one has been freebase.”
  • It wasn’t baseless euphoria like you would get from stimulants. It wasn’t an opiate-like bliss either. It was the kind of bliss that you would get after a nice session of meditation.
  • 40mg/d: I have control over how I want to come off with the blink of an eye. I personally found it almost impossible to get out of bed… you’ll be back to normal within 48 hours.
  • I paired it with PRL-8-53 for weeks and it just made me a machine. However, PRL does not have the same effect on me much anymore. It was very noticeable when I paired it with NSI.
  • Increased appetite & lowered libido
  • I find myself having so many new thoughts in my head that it becomes overwhelming at times and I’ve had to develop new, “better” mental hygiene tactics to deal with the influx of information I seem to be aware of. I have actually been able to reign-in my thought process both in the moment and post facto to use the increased input/output to my advantage.
  • Noradrenaline release in the Hippocampus triggers Neurogenesis, which is a main mechanism of SSRIs, as blocking this has been shown to prevent their antidepressive action.
  • It made me care less about important things, I became very irresponsible.
  • 50mg freebase sublingual: reduces energy to the point I start to need light stimulants just to function throughout the day. Slight anxiety and sociophobia.
  • 50mg freebase: *my vision being “zoomed out” by 5%, like having a slightly larger field of view. Improved memory is the strong point of this substance. I find myself having vivid recollection of memories going back to my single digit ages. Somewhat immature initial emotional responses. For example, jealousy - on a very childish level. *
  • I found it harder and harder to focus as the weeks wore on. All BDNF+ drugs seem to have that issue for those predisposed to lack of focus.
  • Debonaire_death: 5mg is great, >10mg causes anxiety, especially freebase. Many people report it makes them feel like a kid, probably due to BDNF - this effect is characteristic of other substances, too. It makes anything very captivating. However, working memory and sensory gating seem to suffer significantly. Stopping for a few days will revert this whole deal though. Significantly increases sensitivity to racetams, which it is sort of contraindicated with.
  • Be careful mixing it with anything that messes with blood pressure or cortisol. My theory on how NSI-189 works: through the modulation of mineralocorticoids. I first developed the theory because NSI consistently increases racetam sensitivity (he’s talking 100mg here brought things to the negative). NA-Epitalon-Amidate+NSI-189+Sarcosine+Emoxypine cycle, I’ve become hypersensitive to marijuana, even getting uncomfortable effects from it, like parasthesia, and obvious sensations of overstimulation. My memory and logical reasoning are top notch for me right now, though. Something simple L-Theanine or AFA algae extract would do the trick for anxiety.

  • Freebase (~40mg) is better sublingually (and generally more popular),

  • Phosphate (~20-40mg) is better orally and apparently tastes very bad.

    • Yeah there’s the anecdote of a guy going from sedated to stimulated af when he switched to freebase, but there’s also orwello who became an amazing human being all 4 weeks with 10-20 P, 30 P, 40 F, 40 F.
      • I wonder if it’s food when orally administering phosphate inhibiting full absoprtion that gives people potential problems.
    • Bitchyblond has always done 40mg phosphate but saw non-linear improvements on a whopping 80
  • It’s worth noting that it probably achieves some type of saturation, such that a threshold dose may be necessary - generally accepted it’s around 40mg - lest you have no effects, or even adverse effects.

  • Time: Seems it’s personal. Plenty of people take it multiple times per day for it, but in the long term, you can take one dose a week apparently

  • Half-life: Apparently 16+ hours, but acute effects (seems to be borne of adrenals) last 4-6 hours.
