- PARP enzymes initiate mtDNA repair mechanisms, using NAD+ as an energy source. High levels of mice cardiomyocyte mtDNA damage cause a reduction in NAD+ levels due to extreme DNA repair activity, impairing activation of SIRT3.
- Myocardial mtDNA damage combined with high doses of Nicotinamide Riboside causes inhibition of Sirtuin activity due to accumulation of Niacinamide, in addition to irregular cadiac mitochondrial morphology.
- “We show that Nicotinamide Riboside, particularly at high doses, had the opposite effects in cardiac tissue potentially secondary to enhanced NAM evels that would inhibit SIRT3 activity.”
