ALCAR (Acetyl-L-Carnitine)

- Upregulates D1 without acute dopamine increase or D2 upregulation.
- Possibly due to the fact it increases BDNF->ERK.
- ALCAR upregulating D1 increases PKA→CREB, which seems meh at first but apparently ALCAR does NOT activate ΔFosB or CDK5! Thus, it’s just CREB->BDNF, TH, etc., and all the good stuff! How the hell?
- Repeated acetyl-l-carnitine administration increases phospho-Thr34 DARPP-32 levels and antagonizes cocaine-induced increase in Cdk5 and phospho-Thr75 DARPP-32 levels in rat striatum - ’nuff said in title. Persistent after at least 7 days post-treatment as well.
- Seems legit but it’s potently anti-thyroid, no? It’s not high on the to-do list.
Cholinergic, antioxidant.
- Increases mGluR production in the brain (though not hippocampus).
- Increase Choline Acetyltransferase activity in Striatum/Hippocampus. [R](Stimulation of choline acetyl transferase activity by l- and d-carnitine in brain areas of neonate rats)
- Mitochondrial acetylcarnitine provides acetyl groups for nuclear histone acetylation - basically allows it to oppose the actions of HDAC.
