Dutasteride

Dutasteride 180 #

  • Some people inject it like every month/few times a year or something (mesotherapy).

    • The crazy thing is that topically, it doesn’t go as systemic as Finasteride. Will have to read into this more, as it might be Mustang who spread this meme, by doing a scalp biopsy
      • This is because it’s >500 daltons.
      • Same as spironactolone in that they have high molecular weights compared to RU or finasteride.
  • Half-life is 4-6 weeks. It requires 5-6 months to reach steady-state concentrations or to leave the system fully.
  • CYP3A4 and CYP3A5 → 4’-hydroxydutasteride and 6-hydroxydutasteride. CYP4A4 → 15-hydroxydutasteride.
  • Look at all those fluorines, good lord. Increases lipophilicity, half-life, and BBB permeability.

    • Dutasteride reduces serum DHT levels by around 95%,\8]) however at the scalp it appears to only reduce DHT levels by 51% when taken orally at 0.5mg / day.\9]) This would suggest that around 45% of the DHT at the scalp is being produced locally.
      • … then I would suspect at least 50% of the androgenic signalling at our hair follicles is actually coming from locally synthesised DHT. This would suggest that if we can ensure enough Dutasteride is reaching our follicles then it should in theory inhibit 95%+ of the local DHT synthesis like it does in other organs.
    • I also think this opens the door to a highly efficacious combination therapy of a very low dose of oral Dutasteride (perhaps 0.5mg / 14 days) coupled with frequent (preferably daily) topical application which should get us to that sweet spot of 60% - 70% scalp DHT inhibition where progression of androgenic alopecia is rarely ever seen with a very low risk of side effects since when dosed correctly serum DHT should only be reduced by 20% - 30%.

      • (The unabsorbed thing isn’t weirdbtw . 50mg/mL minoxidil only has ~4mg absorbed over 72 hours.)
  • Haircafe - Topical dutasteride. Does it work?
    • So basically apparently it did indeed penetrate into pig skin where the hair follicles are and did not reach the bloodstream. But no other markers were measured. This was almost 4 years ago and not much since?
      • Superior tissue retention = less systemic absorption, so the logic goes. Nanoparticle delivery vehicles such as this should be much less with
